Annealing is a very important process in metalsmithing. When making jewelry, the metal takes a beating, literally! Whether it’s hammered, flattened in a rolling mill, bent, twisted, stretched… the same thing happens, the metal stiffens up and isn’t malleable. This is called “work hardened”. If a metalsmith isn’t careful or paying attention, continuing to work on the piece will result in cracks or it breaking apart. This is where annealing comes in. Annealing is essentially applying heat to a metal so it becomes more malleable again. This process is actually changing the structure of the MOLECULES In the metal. The squished or stretched molecules get reset back to a neutral, unstressed state. This makes the piece of metal malleable again. How cool is that?! PCR, or Polymerase Chain Reaction, amplifies a specific DNA sequence in a series of three cycles with precise temperature control: 1. Denaturation (94-98°C): This is the hottest step, lasting for about 15-30 seconds. Here's what happens: The reaction mixture is heated to a high temperature (typically 94-98°C). At this temperature, the hydrogen bonds between the two strands of DNA in the template DNA molecule break apart, separating them into single strands. 2. Annealing (50-65°C): This stage cools the mixture down to a lower temperature (typically 50-65°C) for 15-30 seconds. Here's why: Short, single-stranded DNA molecules called primers, complementary to the target DNA sequence, are introduced. The cooler temperature allows these primers to anneal (attach) specifically to their complementary sequences on the separated single DNA strands. 3. Extension (72°C): This is the final stage and holds the temperature at a constant 72°C for about 30-60 seconds. Here's what happens: An enzyme called DNA polymerase (often Taq polymerase, named after the Thermus aquaticus bacteria) extends the primers using free-floating nucleotides (building blocks of DNA) present in the mixture. The DNA polymerase adds complementary nucleotides to the 3' end of each primer, synthesizing a new DNA strand complementary to the target sequence. 🧬These three steps are repeated for 25-35 cycles, with each cycle exponentially increasing the amount of the target DNA fragment.
