H & E Staining – Step by Step Procedure 1️⃣ Deparaffinization Slide को Xylene I – 5 min Xylene II – 5 min ➡️ इससे paraffin wax हट जाता है। 2️⃣ Hydration (Alcohol series) Absolute alcohol – 2 min 95% alcohol – 2 min 70% alcohol – 2 min Running tap water – 2 min 3️⃣ Hematoxylin staining Slide को Hematoxylin solution में 5–10 min रखें। ➡️ Nucleus blue/purple हो जाता है। 4️⃣ Washing Running tap water – 5 min 5️⃣ Differentiation 1% Acid alcohol – few seconds ➡️ Extra stain हटाने के लिए। 6️⃣ Bluing Running tap water / ammonia water – 2–3 min ➡️ Nucleus clear blue हो जाता है। 7️⃣ Counterstain (Eosin) Eosin solution – 1–2 min ➡️ Cytoplasm pink हो जाता है। 8️⃣ Dehydration 70% alcohol – few seconds 95% alcohol – few seconds Absolute alcohol – 1 min 9️⃣ Clearing Xylene I – 2 min Xylene II – 2 min 🔟 Mounting DPX mounting medium डालकर coverslip लगा दें। ✅ Result (Microscopy): Nucleus – Blue/Purple Cytoplasm – Pink RBC – Bright pink/red #H&EStaining #Staining #Hematoxlyin #eosin #procedure

It conditions, stains, and seals wood in a single step. No more worrying about pre-stain conditioner, and you only have to sand to 120 grit (yes, really! The can says so!). What makes it foolproof: • It soaks into the wood for buttery, matte color • No lap marks, even on big dresser tops • No “sticky topcoat” drama: the built in sealer cures right with the stain • You don’t need a perfect sanding job (and honestly, it’s more forgiving on open grain) • Application is so fast: I use an edger pad that glides on smooth, covers the whole top without constant re-dipping, and leaves zero brush marks 😍 How I use it: 1️⃣ Sand to 120 grit 2️⃣ Apply stain + finishing oil generously with the edger 3️⃣ Let it sit for 15–20 minutes 4️⃣ Wipe back with a microfiber cloth (trust me, microfiber is better than old t-shirts 😉) 5️⃣ Let it cure. If your end grain always looks darker than the rest of your project, it’s not your stain. It’s your sanding. End grain is a bundle of exposed wood fibers, think bundle of straws, so instead of absorbing stain evenly like the face grain, it soaks it up way more. This is why it turns darker, it’s absorbing more stain. To counter that, sand your end grain to a higher grit than the rest of your board. You might need to play around with your specific wood to see what matches the best. I sand wood to 220 when staining and sand all the way to 600 on the end grain. Hit your grits in between! When you sand to a higher grit, you’re closing up those fibers so they can’t absorb as much stain. Less absorption equals a more even color match. It’s a small step, but it makes a huge difference in the final look whenever ends are visible.
