You’re not crazy, you just have a COMT gene mutation. The COMT gene (catechol-O-methyltransferase) provides instructions for making an enzyme that breaks down certain chemicals in the brain — especially the neurotransmitters dopamine, epinephrine (adrenaline), and norepinephrine (noradrenaline). These neurotransmitters are important for mood, stress response, focus, and emotional regulation. There’s a common variation (or “polymorphism”) in the COMT gene called Val158Met. Everyone has two copies of this gene, and there are three possible combinations: • Val/Val – fast COMT enzyme • Val/Met – medium-speed enzyme • Met/Met – slow COMT enzyme This “speed” refers to how quickly the COMT enzyme clears dopamine and other stress-related neurotransmitters from the brain. Slow COMT (Met/Met) = Breaks down dopamine slowly = Higher dopamine levels, especially in the prefrontal cortex (PFC) = Can lead to increased sensitivity to stress, anxiety, rumination, or even overthinking Fast COMT (Val/Val) = Breaks down dopamine quickly = Lower dopamine levels in the PFC = Can result in focus issues, impulsivity, or difficulty with executive function, but potentially lower anxiety Knowing if you have a COMT gene mutation can help because it gives insight into how your brain handles stress, emotions, focus, and certain medications/supplements. There’s also something called the COMT gene mutation, and what the COMT gene is responsible for is kind of holding those dopamine levels at a good amount in your brain. And so someone might have a slow COMT, and in that case, the dopamine levels can actually raise too much in the brain and cause irritability and anxiety, or someone might have a really fast COMT gene mutation, and that means that somebody’s just cycling through that dopamine really quickly, and they’re gonna feel worse mood-wise. So it’s kind of connected to the MTHFR, where you have the MTHFR pathway connected to the COMT pathway. So both of those things are really crucial to evaluate, especially if you’re considering supplementing with either amino acids or maybe some magnesium.
